Flocytometry protocols for human GIT cell lines

Q1. Can anyone direct me to a good source of protocols on cytotoxicity
studies on human cell lines of the gastrointestinal tract (GIT), especially
those using bombadment with potentially toxic proteins.

Q2. When standardizing substances lethal against bacteria, such as
proteins, for evaluation on human cells, what units of activity among the
following list are most appropriate; arbitrary units/ml, protein
concentration/ml, or LD50 values?. LD50 values being concentations that
kill 50% of experimental bacteria.
