Intracellular cytokines in monocytes

Our group would like to start to assessing cytokine production in
monocytes/macrophages in clinical samples. We have heard some rumors that
measurement of intracellular cytokines in monocytes/macrophages is
problematic but have not heard any specifics. We would appreciate comments
that would help clarify this issue and advise regarding bringing up this
assay.
