The CD41 antigen is the integrin aIIb chain, also called platelet gpIIb, which is non-covalently associated with the integrin b3 chain also called gpIIIa (CD61). Together CD41 and CD61 form the gpIIbIIIa complex. This complex is known as the fibrinogen, fibronectin, vWF, vitronectin and thrombospondin receptor. It is expressed by platelets and megakaryocytes and is involved in platelet aggregation. The gpIIbIIIa is absent or strongly reduced in Glanzmann's thrombasthenia. The P2 antibody reacts with gpIIb in the intact complex with gpIIIa. This antibody blocks binding to fibrinogen and inhibits platelet aggregation induced by thrombin, collagen and ADP. SZ22 antibody reacts with gpIIb independently of the presence of gpIIIa. It is found on platelets and megakaryocytes. This monoclonal does not inhibit platelet aggregation and secretion induced by collagen, arachidonic acid, and thrombin. It does not inhibit fibrinogen binding to human platelets induced by ADP, arachidonic acid and PAF. These three antibodies were evaluated during the Fifth International Workshop on Human Leukocyte Differentiation Antigens.
