May I suggest that you look at the following paper:

Flow cytometric quantification of electroporation-mediated uptake of
macromolecules into plant protoplasts.
Blackhall N.W., Finch R.P., Power J.B., Cocking E.C. and Davey M.R.
Protoplasma (1995) 186:50-56

This paper looks at the uptake of FITC-Dextran and uses PI to assess
viability. In this work we were able to correlate dextran uptake
with transient gene expression following uptake using the same
parameters.
