Our current method is Farahat (1994) J.Clin.Pathol.47,843-849
Basically this is an equal mixture of 4% PFA and BD's
lysing solution for ten mins, spin and resuspend in 0.5%
Tween 20 for 2 mins.
This gives good results for TdT and IgM.
The problem appears to be non specific binding
of anti-MPO-fitc (Dako).
Any comments on other anti-MPO-fitc conjugates,
blocking protocols or methods would be welcome.
