mRNA detection using flow

Hi, we are new to the world of flow, cytokines and RNA and need some help.
We would like to look at cytokine mRNA levels in peripheral blood samples.
Has anyone out there used fluorescent labelled probes to detect mRNA levels
in peripheral blood for detection by flow cytometry (I assume it is
possible to probe for different mRNA concurrently using different fluoro
labels on probes??). Any references or hints would be greatly appreciated
(eg. are there detrimental effects from fixation, how to permeabilise, lots
of REALLY BASIC intructions required!!!).
