cell cycle analysis with PI

Flow'ers
We have started a project looking at intracellular proteins (p21,
p27) in combination with DNA content. Proteins are measured by
indirect fluorescence (FITC GAM) and DNA with PI.

Although our early data is interpretable, we have noticed that the PI
cvs vary and are not always as sharp as we would like.

What factors most affect the tightness of DNA peaks measured with PI?
