In this kit cell mediated cytotoxicity is assessed by flow
cytometric measurement of living target cells
prelabelled with DiOC18 vs. PI-staining dead cells, so
that you can discriminate dead effector cells (only PI
positive) from dead target cells (double positive for
DiOC18 and PI).
I read the reference they cited as literature (J Immunol Meth
156, 47 (1992)), but as a result of this paper you can't use
this method for FACS analysis as cell aggregates between effector
and target cells will make interpretation of the results
impossible.
So my question is if anybody here has some "real life"
experience with this kit, especially regarding a reliable
discrimination of cell aggregates from single cells
and sensitivity in comparison with the 51-Cr release assay.
