Some help for a first time user

I am isolating colonic epithelium from biopsies for flow cytometry.
I am using a simple aspiration technique to help disaggregate the
cells into a partial single cell suspension (5-10% clumping), and
then filter this to remove clumps. However I am concerned about the
amount of debris these 2 procedures generate. Anything up to
50% of the material under the microscope can be cell fragments. Does
anyone have any ideas about cleaning this up? or alternatively a
system which decreases debris production? I would be most grateful
for some advice.
