Ca flux in adherent cells

Need I say more? A visiting investigator wishes to modulate Ca flux
in an adherent cell population. As a pilot, he had tried loading the cells
with Indo while they were still attached, trypsinizing the cells and then
"hitting" them with ionomycin. This was unsuccessful. For a second
attempt, he tried a reagent (that was suggested over this bulletin board-
for use with endothelial cells) which he said gently removed the cells, then
loaded with indo and then tried stimulating with an agent that he has
demonstrated is in fact effective for these cells. He was still unsuccessful.
The question is, can Ca flux be done on adherent cells and then
passed through the flow?
