Steve: No, not me! We have been looking at fish leukocytes only from the
standpoint of DNA content with P.I. I might pass along, however, that we
routinely use DAPI to stain oyster larvae DNA. We get very good histograms
with it, no change in staining characteristics with time (several hours for
each run of around 70-80 samples. The oyster sperm we use for a standard,
remains rock solid in as far as the fluorescence does not diminish or
increase with time. One thing we always do, however, is to allow the sample
to run about 1.0 minute before data collection--just to ensure that
everything stabilizes and their is no inadvertant "leaching" of the dye into
the plastic tubing. It also helps to tap the machine with a "magic wand"
before you begin!

Thanks to all who responded re: my weird DAPI observation--sometimes
I can be amazingly dense (smacking forehead w/ palm).

I would normally check the archive, but my browser's broken-- someone
in this group mentioned antibodies against fish (piscine)
leukocytes--Ross Longley maybe? Could you drop me a line if you know
anything about the subject?
