One of our chaps here is trying to find an appropriate method for
fixing & permeabilising mouse B-cells for analysis of apoptosis, initially
using PI, but after staining for surface antigens. Hopefully, the idea is
to determine the levels of cell death in a cell subset which has been
transfected with a MHC Class-II transgene. Thus far, attempts at fixation
with formaldehyde & with 70% ethanol have resulted in the loss of surface
antigen staining, while B-D tell us that their "FACSlyse" reagent works
with human cells only.
Helpful suggestions welcome (also any further leads on potential
future positions for me [Ph.D. microbiology, plus eight subsequent years
of flow cytometry experience], as I remain on the look-out)...
