Help for the TUNEL method for apoptosis

Based on our experience and suggestions to set up the tecnique,the key points
for the best cell recovery during TUNEL method are:
1) Use PBS-0.1% BSA for washing the cells in every step.
2) After 70% ethanol fixation, spin down the cells at 560xg for 6 minutes at R.T.
3) In all the washing steps spin the cells at 390xg for 5 minutes at R.T.
4) After washing, remove the supernatant very carefully. Do not use the vacuum!
Following these suggestions you should be able to recover at least 50%
af your starting cells.
