Hi,
I think FACSort (and some other instruments from Partec, which has patented
this "piezoelectric principle") is the instrument of choice for rare cells
sorting if you expect small numbers of highly purified cells.
First for its simplicity, so everyone can operate it itself and second for
it is a completely closed instrument, so it is easier to maintain sterile
samples. You can set the sample rate to more than 3000 cells/sec. (according
to BD, but I think it is possible to go even higher), the only limit is
the frequency of sorted cell (up to 300/sec.). I have done some rare cells
sorting and it is really a very simple procedure. Anyway, it is worth to
enrich your population of interest on MACS before throwing it to the flow
cytometer.
